Trypsinogen activation was determined by measuring enzymatic activity of trypsin fluorometrically by using Boc‐Glu‐Ala‐Arg‐MCA as the substrate, as described in detail in Materials and Methods. Trypsin levels (pg/ml) were calculated using a standard curve generated by assaying purified trypsin.

When inappropriate activation of trypsinogen occurs in the pancreatic tissue, TAP is liberated into the circulation and peritoneal cavity, cleared by the kidney, and excreted into urine (2). Trypsinogen became activated in macrophages cultured with purified trypsinogen or co-cultured with pancreatic acini and in pancreata of mice with pancreatitis; trypsinogen activation required macrophage endocytosis and expression and activity of CTSB, and was sensitive to pH. Trypsinogen activation occurs with the hydrolysis of a 7-10–amino acid TAP portion of the N-terminal region of the molecule. Removal of TAP allows the inactive trypsinogen molecule to undergo a conformational change resulting in active trypsin.
